Abstract

A total of 250 specimens were collected from  different clinical sources in Baqubah Teaching Hospital and Al-Batoul Hospital in Diyala /Iraq, during the period from 1/10 /2024 to  30 /12 /2024, Current study investigate bacteriological identification  of Staphylococcus aureus, according to standard methods,  the ability of isolates to Biofilms formation, efflux pumps production, detection the genes for (norB) efflux pumps, (fnbA) biofilms, and (gyrB) quorum sensing was detected molecularly, and  study  the effect of Norfloxacin and Curcumin on the expression of these genes.

The results showed that, there were 25 out of 250 (10%) S. aureus positive growth culture results in the following: wound, nasal, ear, burn, and urine: 6 (24%), 4 (16%), 4 (16%),6 (24%) and 5 (20%), respectively. According to the findings, 15 (60%) of the isolates were methicillin-sensitive S. aureus (MSSA), while 10 (40%) were methicillin-resistant S. aureus (MRSA). The results of the investigation showed that treating isolates with sub-MIC curcumin as QSI significantly decreased efflux pump activity and biofilms degree.

According to the study's findings, the reduction in folding values for all genes norB ,gyrB   and  fnbA  genes recorded with significant differences between this value before and after treatment with sub – MIC of  Curcumin , Norfloxacin  and mixture of the two agents .were the reduction value of  folding mean for this three genes after treatment  equal to  (0.432, 1.175, 0.262) , (0.263, 0.341, 0.039) and (0.547, 0.181, 0.009) respectively .
